BPC-157: Why the animal research gets so much attention

BPC-157 is a 15-residue fragment derived from a protein sequence isolated in gastric juice. The research corpus has an unusual shape: a single prolific group in Zagreb has published rodent work on gut, tendon, ligament, muscle, and nerve injury for two decades. Volume is interesting. Independent replication is a different kind of strength. Human trials are a third. Holding those three apart is how you read this literature well.

What a careful reader can take from the animal work

In rats and mice, BPC-157 has been reported to interact with nitric-oxide pathways, VEGF-related angiogenesis, and FAK-paxillin signaling in tendon fibroblasts. Those are mechanistic hypotheses with animal endpoints — legitimate things to study, and a fair reason the compound draws so much attention. They are hypotheses about animals, not finished claims about human tendons.

Human evidence remains sparse, small, and methodologically thin relative to the online conversation. FDA’s compounding materials have pointed at immunogenicity risk, peptide-related impurities, and the absence of a normal human safety database — the gap a Category 2 listing is designed to mark. In 2026 the Pharmacy Compounding Advisory Committee put BPC-157 related bulk substances on the calendar to discuss ulcerative colitis as a nominated use. That is the Agency doing public work. It is regulatory context, not a clinic opening.

A large rodent literature can be genuinely interesting and still leave the human question wide open.
