Four Knobs, and None of Them Is Glucagon

PTT-A is one peptide aimed at GLP-1, GIP, amylin, and calcitonin. Glucagon is not the fourth knob. The muscle story is a company rat abstract — and a TikTok photo of the slide is not a human result.

Four Knobs, and None of Them Is Glucagon

Someone pauses a TikTok on a slide with four colored triangles and a yellow arrow that says the muscle stays. The comments treat that arrow like a result you can order.

Here's the kindly fix. PTT-A is one peptide from Pep2Tango, aimed at four receptors. Glucagon is not one of them. The muscle picture is a rat abstract in a phone camera.

Picture a remote with four knobs. Retatrutide's extra knob is glucagon. This remote skips that knob and uses amylin plus calcitonin instead. One chain. Not four drugs stirred in a cup.

Our house rule is Literature, not lore. The wider ladder is already up: Is the Future of Weight Loss Already Here? A named capsule such as NA-931 is a different object — see Is the Next Weight-Loss Capsule Already Named? If you want the gut-hormone trunk first, start with how GLP-1 changed the conversation.

What are the four knobs?

GLP-1 is the gut hormone you already hear about. GIP is its partner hormone. Amylin is the "I'm full" signal. Calcitonin is the calcium hormone.

PTT-A is built to lean on all four at once. A 2024 company abstract says the chain matched that idea in lab dishes and cut food intake in lean rats. That is identity work. It is not a prescription.

The phone graphic is the same claim in cartoon form. One beaded chain. Four labels. A face covers the middle, and you can still see it is one string. The cover of this brief is that screen photo. It is a company slide in a phone camera, not a human result.

Did the rats lose weight?

In a 2026 company abstract, obese rats on the higher PTT-A dose lost about 19% of body weight in three weeks versus untreated rats. Pep2Tango's own scientists presented it. You are not the rat.

The lower dose, tirzepatide, and a cagrilintide-plus-semaglutide mix each landed near 12% in that same write-up. The mix was given at a much smaller amount, molecule for molecule. That comparison note lives in the abstract. It is not a recipe.

A 2025 abstract, versus tirzepatide only, printed its own decimals. They sit nearby, they are quoted in the sources, and they are not the labels on this chart.

Three weeks in a rat is not a year in you.

On the TikTok photo, the deepest bracket reads about 19%. The other brackets are small type in the teens. A glare shot will not lock those decimals, so they stay off the claim. Use the abstract. A screen photo is not a second paper, and it is not a human result.

Did the muscle stay?

The 2026 abstract says the loss was mostly fat. Cut-up muscle weights and scans showed muscle loss with the comparators, and preserved muscle with PTT-A. No muscle percent is printed there. Still rats. Still the company.

The 2025 abstract does print one. The two big calf muscles — gastrocnemius and soleus — were about 26% lighter with tirzepatide, and not lighter with PTT-A at the same dose. That sentence is about rats. It does not say PTT-A beats tirzepatide in you.

The chart on the phone is titled as if muscle never drops. Stars sit on some comparator bars. A finger covers the right edge. The slide never labels 26%. Don't read a percent off a finger. The slide is preclinical. It is not a scan of a person.

Are the bone and heart arrows measured?

No. Those slides are a mechanism cartoon, not a results table. Colored triangles stand for the four receptors. Arrows point at brown fat, the heart, a slower stomach, the liver, salt and water, calcium handling, bone, even mood.

One yellow arrow says muscle mass is preserved. Cute drawing. Not a scan of you.

A real calcium line is easy to overread. The 2024 abstract says acute tests in lean rodents lowered blood calcium, and blood glucose with it. That is a lab flag on the calcitonin knob. It is not a bone-density win.

The rat write-ups also describe easier insulin handling, lower triglycerides, and kinder liver-fat signals in the animals. No human dose. No human drug levels. No human heart result.

So is this already a drug?

Not on the record you can check today. Coverage of the June 2026 meeting still filed PTT-A with the preclinical talks. A ClinicalTrials.gov search for Pep2Tango on Oct 3, 2026 came back empty.

For how we grade that gap, see how this journal grades evidence. Inspired Labs does not catalog this peptide. Some other research vial is not this chain. See what Research Use Only means. Research Use Only means laboratory research. It is not a TikTok protocol.

The one thing to remember

Four knobs, and none of them is glucagon. One peptide, still in rats. If you remember one thing, the muscle story is a company abstract, not a result with your name on it.

Company abstract, ADA Scientific Sessions. Quote: “PTT-A produced dose-dependent reductions in cumulative food intake and body weight (−12% and −19% vs vehicle at 10 and 30 nmol/kg, respectively), driven by marked fat mass loss with preservation of lean mass. Tirzepatide and CagriSema each reduced body weight by approximately 12% versus vehicle.” Also: inguinal and epididymal fat reduced 36% and 50% vs 25% with tirzepatide or CagriSema; DEXA total fat-mass reduction 84 g (high-dose PTT-A) vs 60 g (either comparator); carcass analysis and MRI showed significant skeletal muscle loss with the comparators and preserved muscle with PTT-A; glucose, insulin, and triglycerides reduced. Doses: PTT-A 10 or 30 nmol/kg; tirzepatide 30 nmol/kg; CagriSema 2 nmol/kg cagrilintide plus 2 nmol/kg semaglutide; 21 days. The two-drug mix is not the same molar dose as the single peptides. Disclosures include S.S. Ghosh (consultant and shareholder, Pep2Tango) and C. Rondinone (employee and shareholder). No PMID (meeting abstract). Rat data, not a human result.

  2. 0285-OR (2025) — PTT-A vs tirzepatide only; this is where 14.3 / 19.2 / 12.1 and ~26% live — Ghosh et al. (2025). Diabetes 2025;74(Supplement 1):85-OR.

Quote: “Treatment with PTT-A at 10 and 30 nmol/kg over 21 days resulted in significant reductions in cumulative FI and BW (14.3±1.52% and 19.2±0.89% vs vehicle, respectively) driven by reductions in fat mass with no loss of muscle mass. Tirzepatide at 30 nmol/kg reduced BW only by 12.1±0.81% vs vehicle with a decrease in both fat and muscle mass. Gastrocnemius and soleus muscle weights were reduced by approximately 26% with Tirzepatide, with no reduction in muscle weight with PTT-A at same dose.” Also describes triglycerides, insulin sensitization, plasma aminotransferase, plasma lipids, hepatic triglycerides, and liver histology vs tirzepatide at equivalent doses. No CagriSema arm. These decimals are not the brackets on the ©2026 TikTok chart. No PMID. Rat data, not a human result.

  3. 03299-OR (2024) — discovery abstract, including acute calcium — Ghosh et al. (2024). Diabetes 2024;73(Supplement 1):299-OR.

Objective names a long-acting agonist for GLP-1, GIP, amylin, and calcitonin receptors. Results: acute studies in lean rodents decreased blood glucose and calcium; lean rats ate less; chronic DIO rats lost fat mass without loss of muscle mass, unlike tirzepatide in that write-up; lowered plasma insulin, alanine aminotransferase, and liver triglycerides. The published results sentence says “potent GIP, GIP, Amylin, and Calcitonin” — GLP-1 is in the title and objective, then missing in that sentence. Treated as a slip. No human bone or calcium safety package. No PMID. Rat and lean-rodent data, not a human result.

Medscape calls tirzepatide and CagriSema “dual and triple agonists.” The abstract does not. That phrase was not used here. All co-authors reported fees from Pep2Tango, per Medscape. Preclinical meeting coverage, not a human result.
